Monitoring of Antibodies Reaction against Francisella Tularensis Using the ELISA Method

Mikrorganismus Francisella tularensis vyvolává závažné onemocnění lidí a zvířat známé pod označením tularémie. Sledování protilátkové odpovědi na infekci způsobenou tímto mikroorganismem, resp. kmenem s označením LVS (live vaccine strain) bylo provedeno sérologickým vyšetřením vzorků krve získaných od imunizovaných myší inbredního kmene BALB/c v intervalu 1 až 21 dní. Pomocí metody ELISA byly stanoveny a vyhodnoceny titry celkových protilátek a dále izotypových tříd IgM a IgG. V práci se diskutuje o možnosti použití metody ELISA pro diagnostiku tularémie v ranných fázích.

Microorganism Francisella tularensis is the etiological agent of tularemia, a serious disease of humans and animals. Serological diagnosis of antibodies titer in mice models was performed by the indirect ELISA. Inbred mice BALB/c were divided into ten intervals and all of them (without a control group) were inoculated with Francisella tularensis LVS. Obtained blood samples or more precisely sera were collected from 1 to 21 days. Antibodies titers of total immunoglobulins and isotypes IgG and IgM were assessed and finally the progress of antibodies against F. tularensis LVS was characterized. Applicability of ELISA for tularemia diagnosis is discussed.
